Who Is Doing The Voice For Optimus Prime In Transformers?
For the 2007 Live Action Movie it will be Peter Cullen.
He was and will be Optimus Prime's voice. He did the voice of Optimus Prime and Ironhide for the original Transformers Series, but has not been in any of the new Works (RID, ARMADA, ENERGON, CYBERTRON, BEAST WARS, BEAST MACHINES). He is also the guy that does the voice for Eeyore on Winnie the Pooh.
As a side note, Hugo Weaving is Megatron, not Frank Welker who was the voice actor for the original series.

Depends on the series.
In the original animated series as well as the upcoming live action film, Optimus Prime is voiced by Peter Cullen.
In 'Robots in Disguise', Optimus Prime is voiced by Neil Kaplan.
In the Unicron Trilogy (Armada, Energon and Cybertron), Optimus Prime is voiced by Garry Chalk, who voiced Optimus Primal in 'Beast Wars' and 'Beast Machines'.
In the upcoming animated series (premiering on Cartoon Network either this fall or next fall), Optimus Prime will be voiced by David Kaye, best known for voicing the Megatrons of Beast Wars, Beast Machines, and the Unicron Trilogy.
For the Japanese series, I have no idea.
Peter Cullen, the same voice from the animated series.
